Fabrication of Cell Patches Using Biodegradable Scaffolds with a Hexagonal Array of Interconnected Pores (SHAIPs).

چکیده
Cell patches are widely used for healing injuries on the surfaces or interfaces of tissues such as those of epidermis and myocardium. Here we report a novel type of porous scaffolds made of poly(D,L-lactic-co-glycolic acid) for fabricating cell patches. The scaffolds have a single layer of spherical pores arranged in a unique hexagonal pattern and are therefore referred to as "scaffolds with a hexagonal array of interconnected pores (SHAIPs)". SHAIPs contain both uniform pores and interconnecting windows that can facilitate the exchange of biomacromolecules, ensure homogeneous cell seeding, and promote cell migration. As a proof-of-concept demonstration, we have created skeletal muscle patches with a thickness of approximately 150 μm using SHAIPs. The myoblasts seeded in the scaffolds maintained high viability and were able to differentiate into multi-nucleated myotubes. Moreover, neovasculature could efficiently develop into the patches upon subcutaneous implantation in vivo.

Scaffolding plays pivotal role in tissue engineering. In this work, a novel processing technique has been developed to create three-dimensional biodegradable polymer scaffolds with well-controlled interconnected spherical pores. Paraffin spheres were fabricated with a dispersion method, and were bonded together through a heat treatment to form a three-dimensional assembly in a mold. Biodegradab...
متن کاملFabrication of Porous Hydroxyapatite-Gelatin Scaffolds Crosslinked by Glutaraldehyde for Bone Tissue Engineering
In this study, to mimic the mineral and organic components of natural bone, hydroxyapatite[HA] and gelatin[GEL] composite scaffolds were prepared using the solvent-casting method combined with a freeze drying process. Glutaraldehyde[GA] was used as a cross linking agent and sodium bisulfite was used as an excess GA discharger. Using this technique, it is possible to produce scaffolds with mecha...

Gelatin is considered as a partially degraded product of collagen and it is a biodegradable polymer which can be used to produce scaffolds for tissue engineering. Three-dimensional, porous gelatin scaffolds were fabricated by thermally induced phase separation and freeze-drying method. Their porous structure and pore size were characterized by scanning electron microscopy. Scaffolds with differ...
